1. What is the best way to draw and fill the rectangle?

2. When the rectangle rotates it losses its sharpness, is there a way to prevent this from happening?

The quickest way would be to use the rectangular marquee tool. There will be some loss of sharpness as it is being drawn at an angle; this happens with all distortion and transformation techniques.

Another method is to use the shape tool set to path mode. The technique works in almost the same way but does not create individual layers and can be scaled up and down with no loss of quality (although you will still get lose degree of sharpness); you can even create a custom shape from the finished artcile for future use.

Create a rectangle with the marquee tool, or the shape tool. Rotate it, and then fill it after you are finished rotating it, and you will get a nice sharp rectangle.
